## What is kisspeptin

Kisspeptin is the protein product of the KISS1 gene, and it works by binding a single receptor called KISS1R (originally named GPR54 — a docking site on the surface of certain brain cells). That receptor sits on the GnRH neurons of the hypothalamus, the deep brain region that governs reproduction. When kisspeptin docks, those neurons fire and release GnRH in pulses, and the rest of the hormonal cascade follows [1].

The finding that anchored the whole field came in 2003. Researchers found that people born with a broken GPR54 gene failed to go through puberty, and mice engineered the same way reproduced the defect exactly [1]. Close behind it: in women with hypothalamic amenorrhea (missed periods caused by stress, low body weight, or heavy training), a continuous infusion of kisspeptin-54 restored the natural rhythm of LH release, roughly tripling pulse frequency and raising the hormone output about six-fold versus placebo [4]. In healthy men, an intravenous bolus of kisspeptin-10 raised LH about three-fold within half an hour, and a higher continuous rate nudged testosterone up modestly [3].

The metabolism-and-bone angle this review is built around is real but smaller. A short kisspeptin infusion in healthy men raised markers of bone formation by about 20% [11], and decades of animal and human work tie kisspeptin neurons to the body's energy state through the hormone leptin [8][9]. A common search is "kisspeptin supplement," so the plain answer belongs here: kisspeptin is not a dietary supplement. It is a peptide hormone studied as an investigational drug in clinical trials, given by injection or nasal spray under medical supervision [7]. It is not sold as a capsule, it is not regulated as a supplement, and no regulator has approved it for any use.
